Dynabeads magnetic cell separation is a gentle, tube-based technology, so there's no need for any columns. All you need is a magnet!
We released a series of DynaMag cell separation magnets a few years ago, which come in different formats for different needs. They can all be used to isolate pure, viable, and functional cells. Our latest magnetic technology for cell separation is designed specifically for working with FACS or flow cytometry tubes...
This new magnet uses all the great features of our existing magnets plus unique, additional features that make working with FACS or flow cytometry tubes even easier, faster, and more reliable.
The DynaMag-5 will hold any tube between 0.5 and 5 ml. The magnet is strong, and the separation is extremely quick. To make sure you capture all of the beads and cells, we recommend waiting 2 minutes.
Here are a couple additional tips on how to use this magnet.
You should always tilt the whole magnet a few times to make sure you don't lose any beads and cells that might be in the cap or cork. If there's any foam at the top of your sample, it might trap some cells, so shake gently to remove them. If you twist the tubes a little on the magnet, the beads at the tube-wall will form a tighter pellet, and you can lift the pellet a bit from the bottom of the tube. This is helpful for resuspension in smaller volumes.
Use your thumb to hold the tube in place while you open the cap or cork. This way you won't disturb the pellet.
Pull the cap towards you, and make sure you don't miss the few drops that might be caught in the cap.
Now you can pipet off or even decant the liquid, whichever you prefer. It's that easy! Just make sure you get rid of all the liquid. If you decant and there are some droplets left, you can use a small piece of paper. So now you can add your buffer, and lift the tube out of the magnet to resuspend the beadpellet. If you prefer, you can leave the tubes in the rack and resuspend up to 8 samples together. It's really easy to lift the rack off and put it back on the magnet. You can do it with one hand, or you can use both hands. Notice how the rack clicks into place. You can buy extra racks as a separate item.
Efficient washing is important, and here's a useful tip: you can get even better purity if you turn the tube 180º a couple of times. This way the beads and the bound cells will move through the buffer, and that's a great way to wash them.
Keep these tips & tricks in mind. And follow the protocol supplied with your specific product. You'll find that isolating pure, viable, and functional cells is both fast and easy when you use Dynabeads and a DynaMag Magnet!
