David A. Sinclair, a prominent figure in the aging research community, has a history of overhyping his research and possibly glorifying unproven products and supplements, including some from companies with his commercial interests. In April 2023, Dr. Sinclair claimed that a gene therapy developed in his lab and promoted by the co-founded company Life Biosciences had restored vision and reversed aging in monkeys. Many people are very skeptical about his results and proposals, including those who call him a snake oil salesman.

Sinclair's research on resveratrol, a naturally occurring polyphenol found largely in the skins of red grapes, came to scientific attention during the mid-1990s as a possible explanation for the "French Paradox." He claimed that a daily intake of 250 to 500 milligrams of resveratrol may considerably improve health and extend longevity. In 2008, he managed to sell resveratrol as a "magic pill" to a British pharmaceutical company, GlaxoSmithKline (GSK), and personally received about $720 million from the sale. Five years later, GSK halted the research because of the liver toxicity of resveratrol and its underwhelming adverse effects.

David Sinclair was not the first person who claimed health benefits of resveratrol. Dipak Kumar Das (1947–2013) published over 500 articles, including 117 articles on resveratrol. In January 2012, the University of Connecticut announced that a review board had found Das guilty of 145 counts of intentional fabrication or falsification of the experimental results used in published papers. In May 2012, Das was fired from his positions at the University of Connecticut Health Center.

He was elected as president of the Academy for Health and Lifespan Research in June 2023 but resigned in March 2024 after a barrage of rebukes from fellow researchers over his groundless claims of "age reversal" in primates and dogs. A systematic review with 21 co-authors concluded that the published evidence is not sufficiently strong to justify a recommendation for the administration of resveratrol to humans beyond the dose that can be obtained from dietary sources. Resveratrol is yet promoted as a supplement for its potential health benefits, including antioxidant and anti-inflammatory properties.

After the failure to confirm the benefits of resveratrol for longevity, David tried to promote pills with a combination of calcium salt of ketoglutaric acid (Ca-AKG), taurine or quercetin, resveratrol, fisetin, and spermidine. Sinclair practices what he preaches by taking resveratrol with yogurt, NMN, metformin, vitamins D & K2, statin drugs, and aspirin (83mg daily). He proposed cutting out gluten, sugar, and meat and eliminating dairy products from a diet for longevity. In conclusion, it is important to note that David Sinclair's research results have not been reproduced to date, and his status as a scientist is questionable.
