Extreme macro: DIY focus stacking build, WeMacro MicroMate stepping motor, illumination, operation

#AmateurMicroscopy
I explain how I built an extreme macro focus stacking microscope and discuss some important considerations to keep in mind. For optics I use a Sony A99, Minolta 80-200 APO and Mitutoyo objectives. The rig is built on a Leitz Ortholux II microscope stand. I explain the operation of the WeMacro MicroMate stepping motor drive used for automated stacking. I demonstrate some subject illumination methods using fiber and LED lights with various diffusers. Finally, I show the automated focus stacking, image capture operation and a finished result.

Chapters:
0:00 Building using a microscope stand
9:10 Optics and camera
12:58 lighting solutions
21:21 WeMacro MicroMate stepping motor interface and timing
30:55 Stacking operation with example

Mike T points out that the 26 x 0.75 thread for the Mitutoyo objectives works because I'm only using a few threads with the thin adapter. The spec is actually 26mm x 36 tpi (mixed metric and english).

Video equipment and software:
Sony RX-100vii, Godox RL60W LED light with Glow 2 x 2 softbox, Rode wireless GO.
Filmora9, OBS studio, and ApowerMirror smartphone mirroring to PC.
