Principle of Salt Mannitol Agar-Isolation of S.aureus (Staphylococcus isolation)
Salt Mannitol Agar Principle (SMA)
SMA is used as a selective media for the isolation of pathogenic Staphylococci. Staphylococci have the
unique ability of growing on a high salt containing media. Mannitol is the fermentable carbohydrate,
fermentation of which leads to acid production, detected by phenol red indicator. Phenol red indicator is yellow
under acidic conditions and pink/red under alkaline conditions.
1. Mannitol fermenting organism (S.aureus) will produce yellow coloured colonies surrounded by yellow zones
2. Mannitol non fermenting organism will form pink/ red coloured colonies surrounded by red-purple zones
