Soxhlet Fat Extraction Method (Step-by-Step Procedure)
Purpose: To determine crude fat content in food samples using a solvent extraction method.
Materials Required
Soxhlet apparatus
Extraction thimble
Round-bottom flask
Condenser
Petroleum ether (or hexane)
Heating mantle
Analytical balance
Oven
Desiccator
Ground food sample
Step 1: Sample Preparation
Grind the food sample finely.
Dry the sample if it contains excess moisture.
Weigh accurately 2–5 g of sample (W₁).
Step 2: Prepare the Extraction Flask
Clean and dry the round-bottom flask.
Weigh the empty flask and record its weight (W₂).
Add about 150–200 mL petroleum ether to the flask.
Step 3: Load the Sample
Place the weighed sample into a cellulose extraction thimble.
Cover the top of the thimble with cotton wool to prevent sample loss.
Step 4: Assemble the Soxhlet Apparatus
Connect the flask, Soxhlet extractor, and condenser.
Place the thimble inside the extractor body.
Ensure all joints are tight.
Step 5: Start Extraction
Turn on the cooling water for the condenser.
Heat the solvent gently using a heating mantle.
The solvent evaporates, condenses in the condenser, and drips onto the sample.
When the extractor chamber fills, it siphons back into the flask carrying dissolved fat.
Step 6: Continue Extraction
Allow extraction to continue for 4–6 hours (or as specified in the method).
Several siphoning cycles occur automatically.
Fat is gradually extracted from the sample into the flask.
Step 7: Recover the Solvent
After extraction, stop heating.
Remove the thimble.
Distill or evaporate the solvent from the flask.
Step 8: Dry the Flask
Place the flask containing extracted fat in an oven at 100–105°C for 30–60 minutes.
Cool in a desiccator.
Step 9: Final Weighing
Weigh the flask with extracted fat (W₃).

Where:
W₁ = Weight of sample (g)
W₂ = Weight of empty flask (g)
W₃ = Weight of flask + extracted fat (g)
Principle
Fat present in the sample is dissolved by a non-polar solvent (petroleum ether/hexane). Continuous extraction removes the fat, which remains in the flask after solvent evaporation. The increase in flask weight represents the fat content of the sample.
