Alignment of the Movable Objective Microscope (MOM)

Two-photon imaging systems rely on accurate beam alignment for optimal performance. In this tutorial, we will walk through the steps of aligning a Sutter two-photon microscope using a special laser alignment tool and custom pinholes for the Sutter Movable Objective Microscope (MOM®). This procedure will center the imaging laser through the optics of the microscope for best imaging performance. While the alignment tool is specific to a Sutter MOM, the general principles apply to the alignment of any multi-photon laser scanning imaging system.
