Lecture 7. Cytotoxicity evaluation

In this lecture, we will discuss possible ways to assess the cytotoxicity of different compounds, namely, nanoparticles. Firstly, we will discuss modern terminology connected with cytotoxicity. What is called apoptosis and necrosis and what other types of cell death. We will discuss in brief light microscopy-based methods and flow cytometry. Then we will in details discuss fluorescent staining concerning live-dead stain; mitochondrial staining; co-localization of nanoparticles with an ordinary microscope and confocal microscope. We also will discuss fluorescent staining-related problems connected with nanoparticles. Additionally, we will discuss approaches to detect DNA damage. Next, we will move to metabolic or colorimetric assays and how they match fluorescent staining. There will be a couple of examples of why metabolic assay sometimes can provide false information in the real situation in cell systems.
