This mass is based on Josquin's 6 voice motet: Praeter rerum seriem which is scored for SATTBB. This famous motet is based on the Christmas plainchant Praeter rerum seriem. Cipriano de Rore substituted one of the tenors for soprano, and then added a 7th voice, an Alto cantus firmus which sings in very long note values the first three lines of the plainchant melody (Praeter rerum seriem / Parit Deum hominem / Virgo mater) starting either on D or G on the text: Hercules secundus, dux Ferrariae quartus vivit et vivet. (Ercole II, 4th Duke of Ferrara lives and will live.), an hommage to Cipriano de Rore's patron, Duke Ercole II of Ferrara.

This cantus firmus always enters later during the movement except in the Agnus Dei where it starts simultaneously with the other 6 voices.
