Poster - AI-Guided Laser Processing of hiPSC-Cardiomyocytes

To increase the efficiency and reduce cost of human induced pluripotent stem cell derived cardiomyocyte (hiPSC-CM) production we have developed a new artificial intelligence (AI) guided laser processing approach, relying on innovative technology from Kataoka Corp. (Kyoto, Japan). AI was trained to discriminate between cardiomyocytes (CM) and non-CM using CM specific GFP expression on days 8 to 12 of cardiac directed differentiation. For AI training GFP images were overlaid with corresponding phase contrast images to teach AI to identify CMs using phase contrast images only. Now AI can detect CMs using phase contrast images and subsequently commands
laser to ablate non-CMs. After laser processing to ablate non-CMs, the processed CMs were validated using a multi-faceted approach.
