This video describes mechanism of action of spironolactone. It's an aldosterone antagonist. Aldosterone regulates Na reabsorption by principle cells at collecting duct. It increases synthesis as well as activity of Na-K ATPase pump and epithelial Na channels which plays important role in Na reabsorption at collecting duct. Due their increased activity Na reabsorption is increased. Increased Na reabsorption indirectly increases K excretion. Spironolactone inhibits mineralocorticoid receptors through which aldosterone exerts it's effect. So all the effects of aldosterone is decreased. Na is lost in urine and K is prevent from excretion.
