Chaperone-Mediated Autophagy (CMA) | Selection, Mechanism and Regulation

Lesson on chaperone-mediated autophagy (CMA). CMA is a selective degradation process involving the use of protein chaperones, which target and deliver KFERQ-containing protein cargo to the lysosome for degradation. The chaperone complex in CMA consists of a variety of proteins including Hsc70 and Hsp90, which have important functions for CMA. CMA function requires not only proper chaperone functioning, but also lysosomal functioning. Decreased lysosomal functioning due to decreased lysosomal acidification, results in suppression of CMA. When the CMA chaperone complex delivers the protein cargo to the lysosome, the protein cargo is bound, unfolded and then translocated through a lysosomal membrane receptor known as LAMP2A. The protein cargo is then degraded within the lysosome via proteases known as cathepsins. LAMP2A is indispensable for CMA functioning and a deficiency of LAMP2A can lead to a condition known as Danon disease.

In this lesson, you will learn the differences between CMA and macroautophagy, when CMA is active and inactive, and how CMA “selects” its protein cargo through the KFERQ consensus sequence and what the KFERQ sequence is. You will also learn about the roles and purposes of each of the proteins in the chaperone complex, and you will learn the importance and role of various lysosomal proteins in CMA functioning. Finally, you will briefly learn about the genetic condition known as Danon disease, which is due to a deficiency in LAMP2A receptor.
