Joker's father and his sister, Hilary, live in the human colony of Tiptree. After Priority: Thessia, Joker lets Shepard know that the colony was invaded by Reapers several weeks earlier. He hasn't heard any news from his family since the attack started. Liara will later update Joker about the evacuation of Tiptree - some ships carrying refugees, mostly children, recently landed on Salarian colonies. She wasn't able to provide names but this information gave Joker hope that his sister might be alive out there somewhere.

We can learn more about the fate of Joker's sister from a conversation between two Asari in Huerta Memorial Hospital. An Asari huntress named Aeian T'Goni is undergoing psychotherapy on The Citadel, after surviving a reaper attack on a small human colony. Over the course of eight separate conversations, we learn the details of how the Asari huntress survived the attack.

The Asari huntress managed to escape the initial attack, along with a 15 year old human "farm girl," but had to return to the farm to radio for help. When they went back, they discovered indoctrinated human prisoners and the Asari killed them to defend herself and the girl (named Hilary). They managed to get away and hide in a barn but Hilary broke her leg and was unable to stay silent through the pain. Fearing that Hilary’s crying would alert the banshee, the Asari huntress killed her and managed to stay out of sight until she could be evacuated. She blamed herself for not being able to defend the colonists because she didn’t have her gun with her when the attack started.

The girl that Aeian killed was Joker's sister Hilary.

This is an all-around tragic story. Joker lost his family and wasn’t able to say goodbye and he didn’t even know that it happened. The Asari huntress was trying to survive but took some extreme actions to defend herself against the Reapers. She was clearly horrified by her own actions and driven to suicide, if given the chance.
