Sekiro Shadows Die Twice has 40 Prayer Bead Locations to fully upgrade your vitality (health) and posture and get Peak Physical Strength trophy / achievement.
This guide shows all Sekiro Prayer Bead Locations in chronological order to max out your stats.

MISSABLE WARNING: YOU MUST GO FOR THE "PURIFICATION" ENDING TO GET ALL 40 PRAYER BEADS IN ONE PLAYTHROUGH!
If you go for any other ending you'll miss out on some prayer beads (in that case they can only be obtained in New Game+).

Furthermore, after you killed the "Corrupted Monk" main boss in Ashina Depths and "Guardian Ape" in Sunken Valley some of the bosses go away in Ashina Castle. Later on more bosses can disappear so it is vital that you get each of them on the first visit to guarantee everything in one playthrough.

Prayer Beads are collected in 3 ways: located in the open world, dropped by minibosses, bought from merchants.
