In today’s video we will introduce you to the flowcytometry basics,flowcytometry technique, flow cytometry basics principles and how to read flow plots. Flowcytometry is used to count and analyse the size, shape and properties of individual cells within a heterogeneous population of cells. Flow cytometry data is extremely competitive and can be analysed in depth by specific flow cytometry software programmes.
Once a suspension of individual cells is prepared , it is placed on the flow cytometer. How does a flow cytometer analyse the shape, size and other properties of individual cells? What are the different key components of a flow cytometer?
We split the term flowcytometry into three parts, Flow means - in motion, Cyto ~ cell and Metry stands to measure.

So, flow cytometry is the measurement of numerous cell properties as the cells move in a single file in a fluid column while being interrupted by a beam of light.
Our target in flow cytometry is evaluation of properties of single cell. Hydrodynamic focusing is the key to this.
An overview of flow cytometry machine: - Cells in suspension flow in single file past a focused laser where they scatter light and emit fluorescence. Emitted fluorescent light is then filtered by lenses and collected on receptors which convert them to digitized values in the form of plots.

Apart from diagnosing leukemias and hematological malignancies there are few other clinical applications of flow cytometry which include identifing normal hematopoiesis to define the immunophenotypic signature. Staging of lymphomas, minimal residual disease detection , platelet function analysis, studying erythropoiesis and red cell disorders, study of intracellular cytokines, MHC complex analysis, analysis of phagocytosis and cell cycle analysis and detection of apoptosis.
